The objective of this work was to assess the potential of three isolates of arbuscular mycorrhizal fungi to promote growth of micropropagated plantlets of Tapeinochilos ananassae during acclimatization. The experiment was carried out in greenhouse, in a completely randomized block design, with four inoculation treatments: non‑inoculated control and plants inoculated with Glomus etunicatum, Acaulospora longula or Gigaspora albida, with ten replicates. After 90 days, the following parameters were evaluated: survival rate, height, leaf and tiller number, leaf area, fresh and dry biomass, contents of macro‑ and micronutrients in the root and shoot, glomerospore number, and mycorrhizal colonization. The survival percentage was 100%, except for plants inoculated... |

No previous studies have investigated the use of graphite instead of activated charcoal in orchids. In this work, different concentrations of activated charcoal or graphite were added to KC medium to darken the culture medium and stimulate the in vitro propagation of Cattleya bicolor and of a double hybrid orchid (‘BLC Pastoral Innocence’). The seedlings were inoculated on growth regulator-free KC medium; the effects of activated charcoal (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) and graphite (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) concentrations were tested separately. Each flask contained fifteen seedlings, and the experiments had a randomised, two-way factorial design. The two tested variables were culture medium and illumination levels; eleven culture... |

A robust protocol for culturing meristems of Sorghum is required to assist with rapid genetic improvement of the genus. Through meristem culture, an efficient method for rapid micropropagation was developed for Sorghum bicolor (L.) Moench. hybrids, namely NC+262, NC+6C21 and NC+6B50. Complete plants were regenerated directly from shoot meristems without an intervening callus phase. Regeneration frequencies varied between the studied genotypes and according to the growth regulator combinations present in the medium. The combination of BAP and TDZ showed a synergistic effect on shoot multiplication. The highest number of shoots per meristem (68 ± 2) was recorded for the genotype NC+262, followed by NC+6C21 (31.3 ± 0.8), and NC+6B50 (16 ± 1.7). MS medium... |

This study aimed to establish and propagate in vitro plants of Bouchea fluminensis, a medicinal species known in Brazil as gervão-falso ("false verbena"), evaluating the influences of different growth regulators on in vitro multiplication and rooting stages, as well as examining ex vitro acclimatization of rooted plants. Explants were established on Murashige and Skoog medium at half strength of salts and vitamins without growth regulators. For multiplication, the explants were subjected to combinations of 6-benzyladenine (BA; 0, 2.5, 5.0 and 7.5 µM) and α-naphthalene-acetic acid (NAA; 0, 0.2, 0.4 and 0.6 µM). The medium found to induce the greatest number of shoot was that containing 5 µM of BA (NAA-free). For rooting, we evaluated three auxins (NAA,... |

The objective of this work was to characterize the biochemically systemic bacterial isolated from banana plants, to evaluate the bacterial sensitivity to antibiotics, and to determine the phytotoxicity of banana shoots during in vitro proliferation. Systemic bacteria belonging to the Klebsiella and Aeromonas genera were isolated from the “Maravilha” (FHIA 01 AAAB), “Preciosa” (PV 4285 AAAB) and “Thap Maeo” (AAB) varieties and were then characterized. Tests of shoot sensitivity to antibiotics were performed, and the minimum inhibitory concentration (MIC) and phytotoxic effects of selected antibiotics to plants were determined. Among the 20 antibiotics evaluated, the strains showed sensitivity to cefaclor, cefalexin, cefalotin, nalidixic acid,... |

Annonaceae is an ancient family of plants including approximately 50 genera growing worldwide in a quite restricted area with specific agroclimatic requirements. Only few species of this family has been cultivated and exploited commercially and most of them belonging to the genus Annona such as A. muricata, A. squamosa, the hybrid A. cherimola x A. squamosa and specially Annona cherimola: the cherimoya, commercially cultivated in Spain, Chile, California, Florida, México, Australia, Ecuador, Peru, Brazil, New Zealand and several countries in South and Central America. The cherimoya shows a high degree of heterozygosis, and to obtain homogeneous and productive orchards it is necessary to avoid the propagation by seeds of this species. Additionally, the... |

ABSTRACT Multipurpose use of bamboo in rural life makes it as poor man’s timber in Asian countries. Deforestation and industrialization leads to destruction of natural forest. To replenish, a rapid plantation of bamboo could be one of the possible solutions. Bamboo is propagated mainly by vegetative methods though it is not suitable for large scale plantation because of several limitations. Micropropagation is gaining importance for large scale propagation because of its capability in raising huge number of true to type propagules in a limited space in very short span of time. Like any other plant, the chief constraint of bamboo micropropagtion is in vitro contamination arises from several sources including explants. Most of the contaminants are reduced by... |

Croton urucurana Baill., a species belonging to the family Euphorbiaceae, can be useful in the recovery of degraded riparian areas. In the traditional medicine, it is widely used as cicatrizant and in the treatment of rheumatism. However, its seeds present dormancy and low viability, making the propagation of this species a challenge. With the objective of establishing an alternative route for the propagation, the effect of different concentrations of 2,4-D (2,4- dichlorophenoxyacetic acid) with BAP (6-benzylaminopurine) or TDZ (thidiazuron) and of NAA (1-naphthaleneacetic acid) with BAP and their possible combinations were evaluated for callus induction in leaf segments. Callus fresh mass was evaluated forty-five days after inoculation. The isolated use... |

ABSTRACT Nidularium procerum and Nidularium innocentii (Bromeliaceae) were cultivated in vitro on media supplemented with different sources and levels of GA3 (gibberellic acid). These sources were the commercial powder (analytical degree) and fermented extract obtained by Fusarium moniliforme via solid state fermentation. The in vitro elongation and rooting of these plants were evaluated after 50 days of cultivation. The GA3 present in the fermented extract possess the same effect of purified GA3 (analytical degree) for the increase of the height of aerial part of shoots of N. innocentii, but not for the N. procerum being the GA3 fermented extract in a lesser degree. The GA3 fermented extract influences negatively the rooting in N. innocentii, while GA3... |
